Whole cell patch clamp recordings provide basic information about cell firing properties. Recordings are performed using a range of stimulus protocols, including short pulses, long steps, slow ramps, and naturalistic noise to characterize the intrinsic properties of these neurons. This Technical White Paper describes specimen preparation, selection of target regions for experimentation, electrophysiology stimuli strategy, recording and data curation. Slices were prepared either from adult mouse tissue or surgical tissue resected from a human patient. After small differences in the initial part of tissue processing, procedures used for data acquisition and curation for both types of tissue was kept as similar as possible.
